Study on Melting and Mixing Mechanism of Indomethacin in Hot-melt Extrusion

Abstract: The insoluble drug indomethacin (INM) and poloxamer 188 was used as a model drug and a carrier, respectively, and a solid dispersion of indomethacin and poloxamer 188 was prepared by a twin rotor mixer. The effect of rotor speed ratio on the melt transition and mixing process of INM was studied in terms of the two aspects of numerical simulation and experimental investigation. The results indicated that the distribution, dispersion mixing and melt diffusion of INM in the carrier could be enhanced by increasing the rotor speed ratio, and the dissolution process of INM was also accelerated.
